Up-to-date imaging approaches were used to address the spatiotemporal organisation of the endomembrane system in secretory cells of Dionaea muscipula. Different ‘slice and view’ methodologies were performed on resin-embedded samples to finally achieve a 3D reconstruction of the cell architecture, using ultrastructural tomography, array tomography, serial block face-scanning electron microscopy (SBF-SEM), correlation, and volume rendering at the light microscopy level. Observations of cryo-fixed samples by high-pressure freezing revealed changes of the endomembrane system that occur after trap activation and prey digestion. They provide evidence for an original strategy that adapts the secretory machinery to a specific and unique case of stimulated exocytosis in plant cells. A first secretion peak is part of a rapid response to deliver digestive fluids to the cell surface, which delivers the needed stock of digestive materials ‘on site’. The second peak of activity could then be associated with the reconstruction of the Golgi apparatus (GA), endoplasmic reticulum (ER) and vacuolar machinery, in order to prepare for a subsequent round of prey capture. Tubular continuum between ER and Golgi stacks observed on ZIO-impregnated tissues may correspond to an efficient transfer mechanism for lipids and/or proteins, especially for use in rapidly resetting the molecular GA machinery. The occurrence of one vacuolar continuum may permit continuous adjustment of cell homeostasy. The subcellular features of the secretory cells of Dionaea muscipula outline key innovations in the organisation of plant cell compartmentalisation that are used to cope with specific cell needs such as the full use of the GA as a protein factory, and the ability to create protein reservoirs in the periplasmic space. Shape-derived forces of the pleiomorphic vacuole may act as signals to accompany the sorting and entering flows of the cell.  相似文献

A small-scale silica gel-water adsorption system with modular adsorber, which utilizes solar energy to achieve the cogeneration of domestic air conditioning and water heating effect, is proposed and investigated in this paper. A heat recovery process between two adsorbers and a mass recovery process between two evaporators are adopted to improve the overall cooling and heating performance. First, the adsorption system is tested under different modes (different mass recovery, heat recovery, and cogeneration time) to determine the optimal operating conditions. Then, the cogeneration performance of domestic cooling and water heating effect is studied at different heat transfer fluid temperatures. The results show that the optimal time for cogeneration, mass recovery, and heat recovery are 600 s, 40 s, and 40 s, respectively. When the inlet temperature of hot water is around 85°C, the largest cooling power and heating power are 8.25 kW and 21.94 kW, respectively. Under the condition of cooling water temperature of 35°C, the obtained maximum COPc, COPh, and SCP of the system are 0.59, 1.39, and 184.5 W/kg, respectively.  相似文献

Horizontal directional solidification experiments were carried out with a monophasic Sn-2%Sb (mass fraction) alloy to analyze the influence of solidification thermal parameters on the morphology and length scale of the microstructure. Continuous temperature measurements were made during solidification at different positions along the length of the casting and these temperature data were used to determine solidification thermal parameters, including the growth rate (VL) and the cooling rate (TR). High cooling rate cells and dendrites are shown to characterize the microstructure in different regions of the casting, with a reverse dendrite-to-cell transition occurring for TR>5.0 K/s. Cellular (lc) and primary dendrite arm spacings (l1) are determined along the length of the directionally-solidified casting. Experimental growth laws relating lc and l1 to VL and TR are proposed, and a comparative analysis with results from a vertical upward directional solidification experiment is carried out. The influence of morphology and length scale of the microstructure on microhardness is also analyzed.  相似文献

The efficiency of induction ring-type heating systems and on an unclosed magnetic circuit, as applied to soldering parts and electronic components, is sized up during assembly of electronic modules. For inductors on the magnetic circuit, owing to the concentration of the electromagnetic field in a working gap, the efficiency of heating of parts increases and the heating zone is localized. Simulation of the parameters of both the electric and magnetic fields in COMSOL gives the temperature profiles of heating of surfacemounted components over a frequency range of 22–100 kHz. The highest heating rate of 12–15°С/s is attained at lower frequencies, where the penetration of the electromagnetic field is deeper.  相似文献

Nanosized particles of CoAlxFe2-xO4, where 0?≤?x?≤?2, were synthesized by the sol–gel combustion method and the magnetic properties of these compounds were investigated. According to X-ray diffractograms, the samples are single phase and the crystallite size is between 7 and 25?nm. The room temperature saturation magnetization of the samples was estimated from the cation distribution and ferromagnetic resonance spectra were used to determine the magnetocrystalline anisotropy. The results show that the saturation magnetization and the magnetocrystalline anisotropy vary over a wide range, from maxima of Ms =?0.42?MA/m and K?=?0.39?kJ/m3 for x?=?1.0 to minima of almost zero for x?≈?1.4, a result that could be useful for practical applications of these materials.  相似文献

Lean practices are known to increase operational performance. Previous research has identified critical success factors for implementing lean practices. This research aims to examine the extent to which success factors are critical for various degrees of lean practice implementation. Using multiple-respondent self-assessments from 33 Dutch manufacturing small and medium-sized enterprises (SMEs), we conducted a Necessary Condition Analysis. Our findings indicated that the criticality of success factors is progression dependent. In the initial stages of the lean journey, SMEs could improve their lean practices in a bottom-up manner through local factors such as a learning focus, improvement training and support congruence. When lean practices are more advanced, some company-wide factors must be present: top management support, a shared improvement vision and a supplier link. Our findings question the universality of success factors such as strategic involvement and indicate the need for a more dynamic model of lean implementation.  相似文献
